Detecting and rewiring cellular interactomes
Changes in protein localization and abundance are hallmarks of altered cell state and function. We develop technologies that harness mammalian machinery to manipulate and measure these changes, enabling mechanistic insight and therapeutic discovery. Targeted protein relocalization and programmed manipulation of RNA using human Argonaute proteins exemplify recent approaches to directly rewire cellular behavior. In parallel, we program cellular circuits that relay downregulation of a protein-of-interest into user-defined outputs via ratiometric transcriptional activation. These systems provide scalable and sensitive readouts for small molecule discovery and high-throughput mapping of proteostasis networks for reprogramming cell behavior and addressing challenging disease targets.
